目的 基于通关藤抗炎作用,采用“多谱-多效”关联分析筛选其质量标志物。方法 利用高效液相色谱-紫外检测器-蒸发光散射检测器(HPLC-VWD-ELSD)技术,建立通关藤有机酸类、甾体皂苷类多重指纹图谱。检测炎症因子一氧化氮(NO)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)的含量,作为多重抗炎指标评价通关藤对脂多糖(LPS)诱导的RAW264.7细胞抗炎作用。结合灰色关联分析(GRA)、偏最小二乘回归分析(PLSR),通过“多谱-多效”关联筛选质量标志物,并利用分子对接技术验证。结果 建立了含18个共有峰的通关藤多重指纹图谱,并指认了其中12个峰。经GRA、PLSR分析确定了5个通关藤抗炎作用的质量标志物,分别为通关藤皂苷I、H、G和通关藤苦苷B、A,5个标志物共同作用的抗炎核心靶点为MAPK14、AKT1、MAPK1、CASP3。结论 确定了通关藤抗炎作用的质量标志物,为通关藤质量控制和深入开发提供了依据。
Abstract:Objective To identify the quality markers of Marsdenia tenacissima (M.tenacissima) by determining its anti-inflammatory effect and integrating multi-spectrum with multi-effect.Methods Multi-fingerprint patterns of organic acids and steroid saponins in M.tenacissima were established with high-performance liquid chromatography-ultraviolet detector-evaporative light scattering detector (HPLC-VWD-ELSD).Nitric oxide (NO),tumor necrosis factor-α (TNF-α),and interleukin-6 (IL-6) were determined as multiple anti-inflammatory indicators to evaluate the effect of M.tenacissima extracts on LPS-induced RAW264.7 cells.Quality markers were screened by multi-spectrum-multi-effect correlation with grey relational analysis (GRA) and partial least squares regression (PLSR),and verified by molecular docking.Results Multi-fingerprint patterns of 18 common peaks were established for M.tenacissima,12 of which were identified.Through GRA and PLSR,five quality markers of the anti-inflammatory effect of M.tenacissima were determined (tenacissoside I,tenacissoside H,tenacissoside G,marsdenoside B,and marsdenoside A).The core anti-inflammatory targets common to these five markers included MAPK14,AKT1,MAPK1,and CASP3.Conclusion The quality markers of the anti-inflammatory effect of M.tenacissima are identified,providing a basis for quality control and in-depth development of M.tenacissima.
