目的 研究核桃Juglans regia L.青皮乙醇提取物的化学成分及抗炎活性。方法 利用硅胶、葡聚糖凝胶、重结晶等方法分离核桃青皮乙醇提取物的化学成分,通过波谱学方法(MS,NMR)鉴定其化学结构,并测定其脂多糖(LPS)诱导RAW 264.7巨噬细胞释放一氧化氮(NO)水平的影响。结果 从核桃青皮95%乙醇提取物中分离得到15个化合物,分别鉴定为:4,8-二羟基-1-四氢萘酮(1)、4-乙酰氧基-8-羟基-1-四氢萘酮(2)、4-十六酰氧基-8-羟基-1-四氢萘酮(3)、4-乙氧基-1-四氢萘酮-5-O-β-D-吡喃葡萄糖苷(4)、3-羟基乌苏-12-烯(5)、熊果酸(6)、3,4-二甲氧基苯甲酸(7)、3,4-二羟基苯甲酸甲酯(8)、大黄酸(9)、十六烷酸(10)、十六烷酸甲酯(11)、二十二烷醇(12)、乙二醇十六烷酸乙酸酯(13)、β-谷甾醇(14)、胡萝卜苷(15)。结论 化合物2~4和13为首次从胡桃属中分离得到,化合物2~5和8~13为首次从核桃青皮中分离得到,其中化合物9和11可显著抑制NO释放且细胞毒性较低。
Abstract:Objective To determine the chemical constituents and anti-inflammatory activity of alcohol extract of green husk of Juglans regia L..Methods The constituents were isolated and purified by silica gel column chromatography,Sephadex LH-20 column chromatography recrystallization,etc.Their structures were elucidated based on physiochemical evidence and spectral data (MS,1H-NMR,13C-NMR).The anti-inflammatory activity was evaluated by measuring nitric oxide (NO) levels released by lipopolysaccharide (LPS)-induced RAW 264.7 cells.Results Fifteen compounds were isolated from 95% ethanol extract and identified as 4,8-dihydroxy-1-tetralone (1),4-acetoxyl-8-hydroxy-1-tetralone (2),4-hexadecyl-8-hydroxy-1-tetralone (3),4-ethoxy-1-tetralone-5-O-β-D-glucuronopyranoside (4),3-hydroxyursic-12-ene (5),ursolic acid (6),3,4-dimethoxy-benzoic acid (7),methyl 3,4-dihydroxybenzoate (8),rhein (9),hexadecanoic acid (10),methyl hexadecanoate (11),docosanol (12),glycol hexadecanoate acetate (13),β-sitosterol (14),and daucosterol (15).Conclusion Compounds 2~4 and 13 have been isolated from the genus Juglans for the first time and compounds 2~5 and 8~13 from the green husk of Juglans regia L.for the first time.Compounds 9 and 11 obviously inhibit the release of NO with low cytotoxicity.
